ABSTRACT
Genetically-Encoded Glycopeptide Libraries
Genetically-encoded (GE) libraries of proteins and peptides are one of the major sources of discovery of biological drugs and development of ligands. These techniques, however, are often limited to handling of structures made of the 20 natural amino acids. Our group uses GE-libraries of peptides as a starting material for multi-step organic synthesis to produce GE-libraries of peptide derivatives with a specific emphasis on the production of chemically-glycosylated peptide and macrocyclic peptide libraries. Examples are N-terminal conjugation1 and cyclization of linear peptides2 with simultaneous introduction of glycan entities. These chemical modifications allowed us to develop the Genetically-Encoded Fragment-Based Discovery (GE-FBD) platform,3 which combines >109 peptide fragments with variable, silently-encoded modifications.4 The talk will describe the progress and challenges in application of GE-FBD platform to several carbohydrate binding proteins, such as ConA,3 Galectins, DC-SIGN and anti-LAM antibodies. The vision and development of the technology to maximize the reproducibility of discovery within a genetically-encoded library framework will be discussed.
BIO
Ratmir Derda received his B.Sc. in Physics from Moscow Institute of Physics and Technology in 2001. He completed his Ph.D. in Chemistry from the University of Wisconsin-Madison in 2008, under the supervision of Laura L. Kiessling, and postdoctoral training at Harvard University under the supervision of George M. Whitesides and Donald E. Ingber. He joined the University of Alberta in 2011 and in 2017 he was promoted to Associate Professor. Notable awards include the David Gin New Investigator Award (2017), Martha Cook Piper Research Prize (2017), and Rising Star in Chemical Biology (2015).
